UK firm Robert Wiseman Dairies won approval early in 2005 for the huge development.
It is being built on land off Adderley Road, near the town's Gingerbread Man pub.
The site will be used by 50 vehicles to collect milk from more than 400 farms in north Shropshire, north Staffordshire and Cheshire. The Scottish company is holding a recruitment drive and is on the lookout for security staff, drivers, cleaners, laboratory analysts and maintenance and yard workers.
A spokesman said: "Our state-of-the-art site is due to open at the start of June and we are looking for enthusiastic and committed individuals who will play an integral role in supplying more than one billion litres of milk to many of the country's leading supermarkets and convenience retailers."
North Shropshire District Council chiefs approved the new site as they said there would be clear economic benefits for the town and surrounding area.
The plans were backed by Market Drayton economic development forum and Advantage West Midlands.
Graham Jack, commercial director for Robert Wiseman and Sons, said: "The council's economic development and planning support team has been invaluable in helping us achieve our investment in the north Shropshire area.
"We are pleased to be able to offer our £4.5 million pound investment, which has created 100 new jobs."
