Paul Hurst's side will head to Vale Park in the week commencing December 6.
England Under-17 World Cup-winning manager Steve Cooper and Southampton legend Matthew Le Tissier conducted the draw at Wembley.
Town would have had a home draw had they avoided defeat in their final group game against Walsall on Tuesday.
Vale, currently 22nd in League Two, topped a group that also contained Oldham, Newcastle Under-21s and Crewe.
