Ferrell and Rose will again take over from loan stars John Eustace and Neil Mackenzie who have not been given permission to play in the Cup by their clubs.
Both Ferrell and Rose played well in the first meeting and are eager to get back into the action.
"I thought Richard Rose, Tim Sills and myself did well at the Gay Meadow considering we had been out for a long time," said Ferrell.
"We created quite a number of chances and we also defended very well. But we only did half the job, now we have to make sure we do the business at Edgar Street."
Hereford are also without striker Stuart Fleetwood and will have to check on his replacement Tim Sills.
He suffered a head injury in the second half against Walsall on Saturday.
Club captain Rob Purdie, who leads Hereford's goal scoring with six, is looking to the supporters to lift them to victory. "We could not have a bigger incentive to win than a second round tie at home to Port Vale," he added.
