"I'm really looking forward to it and hopefully I can get through the 90 minutes," said the 22-year-old.
"I have been waiting to play for a long time but I went to see the specialist last Thursday and he gave me the all-clear and I don't have to wear a cast or anything like that now.
"I have been able to train with the lads for the last few weeks which was important because I felt in really good shape at the start of the season and I wanted to retain that sharpness.
"Tonight will give a chance for a few of the lads who haven't been in the League team to show what they can do so it's a good game for us to have.
"We beat Yeovil in the same competition last season so hopefully we can do it again."
Manager Gary Peters has hailed Ashton's return, adding: "It's a massive boost for us because it puts Marc Tierney under pressure at left-back, Marc Pugh under pressure on the left of midfield and the two central midfielders under pressure."
Competition rules state at least six of the 11 players who have featured most this season, but Peters is still likely to ring the changes with Stuart Nicholson almost certain to partner Michael Symes in attack.
Elsewhere, Darran Kempson, Chris Humphrey, Steve Leslie and Jimmy Ryan may all be given starts while skipper Stewart Drummond, Glyn Garner and Colin Murdock could be rested.
Tonight's game will go straight to penalties if the scores are tied after 90 minutes.
